Bombus terrestris developmental DNA methylation dynamics

This dataset contains DNA sequence data from reproductive Bombus terrestris audax workers brain and ovarian tissue (80 – 100 days post establishment), and from the tissue of male offspring of these workers at varying developmental stages, specifically stage 4 larval heads, pupal heads, adult (13 – 14 days old) male brains and adult male sperm.

Provenance & quality

DNA was extracted from using the Qiagen DNA Micro Kit following the manufacturers protocol. Each sample was snap-frozen in liquid nitrogen and stored at -80C. DNA from two/three organisms was pooled to create representative samples for each colony. In summary, 20 samples were collected – 4 * reproductive worker brain, 4 * ovary, 4 * larval head, 4 * pupal head, 2 * male brain and 2 * sperm.

DNA quantity and quality were determined by Nanodrop and QubitR fluorometers. DNA was sent to BGI Tech Solution Co., Ltd.(Hong Kong) for sequencing using Illumina HiSeq 4000.

Sequencing data were checked using fastqc v.0.11.5 and adapters and poor quality bases were trimmed using bbmap (
